| 内容記述 | In the experiment, we devised an apparatus for measuring the size of a single callus. That is, the area of a growing callus in silhouette projected on a sheet of plotting paper was measured at intervals and the relative growth rate was calculated. By using this method, the effect of aluminum on growth of a single callus was investigated. And we also examined the acquisition of tolerance in calli and in plantlets produced on calli through somatic embryogenesis. The results showed that the calli which had beam grown on the medium supplemented with high concentrations of aluminum grew more rapidly than the others did, when they were transferred on the medium containing high concentrations of aluminum. It can be said that the calli have acquired aluminum tolerance. On the other hand, the proportion of calli with embryoids was reduced by high concentrations of aluminum. And the percentage of plantlets with normal morphology was also reduced. It is considered that the metabolism involved in cell differentiation is more sensitive to aluminum than that involved in call proliferation is. | |||||
