| 内容記述 | D-Aspartate oxidase (EC 1.4.3.1) was purified 195-fold from pig kidney cortex. The purified preparation had a specific activity of 5279 nmol/min per mg of protein with D-aspartate as substrate and consisted of two major proteins of 53 kDa and 57 kDa with other minor proteins as indicated by sodium dodecyl sulfate polyacrylamide gel electrophoresis. The molecular mass of the native enzyme was estimated to be 230 kDa by gel filtration. The optimum pH was rather broad ranging from 7.5 to 9.5 with a maximum at 8.5. The optimum temperature was 45\C\, while the stability decreased with the increase in temperature above 30\C\. The enzyme did not follow Michaelis-Menten kinetics probably because of substrate activation. For this reason, the Km values were obtained with higher concentrations of substrates and were 4.77, 1.91 and 51.1 mM for D-aspartate, N-menthyl-D-aspartate and D-glutamate respectively. Of the compounds tested meso-tartrate was the most effective competitive inhibitor with a Ki calue of 1.44mM. To utilize the purified preparation for enzymatic analysis of acidic D-amino acids, various reaction conditions were tested. The reaction systems thus established including those for differential determination of D-aspartate and D-glutamate are described. | |||||
